{"id":552,"date":"2008-12-11T22:11:21","date_gmt":"2008-12-11T20:11:21","guid":{"rendered":"http:\/\/blog.ultimatefatburner.com\/?p=552"},"modified":"2015-02-12T21:13:57","modified_gmt":"2015-02-13T02:13:57","slug":"pathetic","status":"publish","type":"post","link":"https:\/\/www.ultimatefatburner.com\/ufb-blog\/pathetic\/","title":{"rendered":"This is Pathetic&#8230;"},"content":{"rendered":"<p>I&#8217;m currently writing a review of LG Sciences pSARM.\u00a0 Evidently, the company is\u00a0doing\u00a0its best to convince would-be customers that the primary ingredient, osthol, binds to androgen receptors.\u00a0 So one of their reps\u00a0<a href=\"http:\/\/anabolicminds.com\/forum\/supplements\/83176-psarm-write-up.html\" target=\"_blank\" rel=\"noopener\">posted a drawing superimposing osthole over testosterone<\/a>\u00a0on a bbing board to\u00a0&#8220;prove&#8221; it &#8220;fits&#8221; androgen receptors.*<\/p>\n<p>There&#8217;s just (ahem!) a few problems with this argument&#8230;<\/p>\n<p>1. Ligands &#8211; and their binding sites &#8211; are <a href=\"http:\/\/web.archive.org\/web\/20110809074324\/http:\/\/users.ox.ac.uk:80\/~mwalter\/web_05\/resources\/chime_web\/conform_analysis.shtml\" target=\"_blank\" rel=\"noopener\">THREE-DIMENSIONAL<\/a> &#8211; not two.\u00a0\u00a0What LG Sciences has done\u00a0is a bit like looking up at the night sky at a constellation, and assuming all the stars in a particular formation lie in the same plane, simply because that&#8217;s what it looks like from Earth.\u00a0 2-D chemical formulas are simply a shorthand way of representing molecules, but like a painting or photograph, have no depth. Seemingly mall differences in structure can add up to large differences in shape &#8211; where it matters.  And despite the alleged similarity, there are some pretty significant differences between the two.<\/p>\n<p>2. If 2-D structural similarity is all that matters, there are plenty of other\u00a0compounds that are a much\u00a0closer match for testosterone\u00a0than osthol is.  Like estradiol (estrogen), for example. \ud83d\ude00<\/p>\n<p>3. If 2-D structural similarity is all that matters, then perhaps the\u00a0brain trust\u00a0at LG Sciences would care to explain why <a href=\"http:\/\/www.ncbi.nlm.nih.gov\/pmc\/articles\/PMC2072879\/table\/T2\/\" target=\"_blank\" rel=\"noopener\">many of the SARM drugs currently being researched<\/a> have even less structural overlap w\/testosterone than osthol does.<\/p>\n<p style=\"text-align: left\">In short,\u00a0it&#8217;s just marketing, and a rather\u00a0weak attempt at\u00a0that.\u00a0\u00a0You &#8220;prove&#8221; androgen receptor binding capacity by &#8211; surprise!!! &#8211; <a href=\"http:\/\/www.ncbi.nlm.nih.gov\/pmc\/articles\/PMC2072875\/?tool=pubmed\" target=\"_blank\" rel=\"noopener\">conducting receptor binding studies<\/a>\u00a0(scroll down to &#8220;In Vitro and In Vivo Models for Evaluation of AR Ligands&#8221;)\u00a0You\u00a0&#8220;prove&#8221;\u00a0a compound acts like a SARM by <a href=\"http:\/\/www.ncbi.nlm.nih.gov\/pubmed\/18502117\" target=\"_blank\" rel=\"noopener\">demonstrating it has tissue-selective effects<\/a>.\u00a0<\/p>\n<p style=\"text-align: left\">If you&#8217;re going to play &#8220;scientist,&#8221; then you need to play by the &#8220;rules.&#8221;\u00a0 It&#8217;s that simple.<\/p>\n<p style=\"text-align: left\">Pathetic.<\/p>\n<p>*This drawing is referred to in the pSARM write up on their web site, but does not actually appear there.
